PENGARUH KONSENTRASI ETANOL DAN LAMA MASERASI TERHADAP KARAKTERISTIK EKSTRAK DAUN KUDA-KUDA (Lannea coromandelica)

Abstract

The plant species Lannea coromandelica is highly valued in Indonesian traditional medRahayu, Tyas Dwiicine for its rich profile of bioactive compounds. This study was systematically designed to optimize the maceration process by examining the simultaneous effect of ethanol solvent concentration (70%, 80%, 90%) and maceration time (24 hours, 48 hours) on extract quality. A Factorial Completely Randomized Design (CRD) with four replications (totaling 24 treatments) was employed. The extracts were analyzed for water content, yield, total phenolic content, antioxidant activity, antibacterial efficacy against S. aureus and E. coli, and phytochemical groups via FTIR analysis. Statistical evaluation was conducted using ANOVA, followed by the Duncan Multiple Range Test (DMRT). Results indicated that increasing ethanol concentration significantly enhanced the extraction yield. FTIR confirmed the presence of crucial secondary metabolites, including alkaloids, flavonoids, terpenoids, saponins, and tannins. While the extract exhibited antibacterial activity against both S. aureus and E. coli, its effectiveness was inferior to the Amoxicillin control. The most effective extraction condition was achieved using 90% ethanol and a 48-hour maceration time. This optimal treatment maximized both the antioxidant activity (86.39%) and the total phenolic content (7.24 mg GAE/g). In conclusion, the best method for obtaining a high-quality Lannea coromandelica extract rich in antioxidant and phenolic compounds is through maceration using 90% ethanol for 48 hours.
